Objective: To transfect KiSS-1 into HEC-1B cell lines in a safe, high-efficiency and low toxicity way, and then to find out whether KiSS-1 affects the metastasis ability of cell lines. Methods: Lipofect was used to transfect KiSS-1 into HEC-1B cell lines. The difference in the expressions of KiSS-1mRNA, MMP-9 mRNA and metastin between the transfected and non-transfected cell lines was detected by RT-PCR and Western blot. Results: 1) By using the system, the stable expression of KiSS-1 mRNA and the significant difference in cells, expression between transfected and non-transfected cell lines were observed. 2) The relative content of KiSS-1 mRNA and metastin protein were 0.1231 ± 0.0366, 0.1486 ± 0.0287, 0.5060 ± 0.0198 and 0.1549 ± 0.0189, 0.1497 ± 0.0029 and 0.4469 ± 0.0078, respectively. There was a significance between the plasmid group and other groups (F = 161.547, P = 0.000; F = 607.993, P = 0.000). 3)The relative contents of MMP-9 mRNA were 0.3576 ± 0.050 1, 0.3500 ± 0.0254 and 0.1677 ± 0.0014, respectively. There was a significance between the plasmid group and other groups (F = 32.963, P = 0.001). Conclusions: The way that transfects KiSS-1 into the HEC-1B cell lines is safe, high-efficient and low toxic. The expression of KiSS-1 is increased and the expression of MMP-9 is decreased significantly in the transfected cell lines. KiSS-1 is a promising candidate tumor-suppressor gene and may play a key role in the metastatic cascade.
