Objective: To study the mRNA expression of K-Cl cotransporter 1 (KCC 1) in endometrial cancer and investigate the effect of silencing KCC 1 gene by siRNA interference on the proliferation and cell cycle of endometrial cancer HEC-1B cells. Methods: Fluorescent quantitative PCR was used to detect the mRNA expression of KCC 1 gene in normal endometrial tissues and endometrial cancer tissues. Specific siRNA for KCC 1 gene was designed and synthesized. The HEC-1B cells were transfected with specific siRNA to silence the expression of KCC 1 gene. After RNA interference, the KCC 1 mRNA and protein expressions were detected using RT-PCR and Western blotting, respectively. MTT assay was used to detect the proliferation ability. FCM was performed to examine cell cycle distribution and apoptosis of HEC-1B cells. Results: The expression of KCC 1 mRNA in endometrial cancer tissues was much higher than that in normal endometrial tissues. The difference has statistical significance (P < 0.05). The KCC 1 specific siRNA markedly inhibited the tissues KCC 1 mRNA and protein expression in HEC-1B cells (P < 0.05). Cell cycle analysis showed that HEC-1B cells were arrested at G0/G1 phase. The apoptotic rate was increased compared with control group (P < 0.05). Conclusion: There is a relationship between KCC 1 gene expression and endometrial cancer. KCC 1 gene was involved in the regulation of cell cycle and had the power to stimulate the proliferation of tumor cells.
